Karakter Morfologis dan Genetik Jamur Tiram (Pleurotus Spp.)

Hutan tropis Indonesia merupakan salah satu pusat keanekaragaman hayati di dunia, yang salah satu di antaranya ialah jamur tiram (Pleurotus spp.). Penelitian ini bertujuan mempelajari karakteristik morfologi dan genetik delapan isolat Pleurotus spp.. Penelitian dilakukan pada bulan September 2005 sampai April 2006 di Laboratorium Patologi Hutan dan Lab. Silvikultur, serta Lab. Bioteknologi Kehutanan dan Mikrobiologi Molekuler, Pusat Studi Hayati, Institut Pertanian Bogor. Tubuh buah delapan isolat jamur digunakan sebagai bahan pengamatan. Hasil penelitian menunjukkan bahwa tubuh buah delapan isolat jamur memiliki warna putih, coklat, atau merah jambu, dengan atau tanpa tangkai, bentuk tudung berupa lingkaran penuh atau setengah lingkaran. Amplifikasi dengan primer RAPD OPO11 menghasilkan 12 pita, satu pita bersifat monomorfik dan 11 pita lainnya bersifat polimorfik yang menunjukkan keragaman pada delapan isolat jamur tiram yang dipelajari. Pengelompokan berdasarkan pola pita amplifikasi primer RAPD tersebut menghasilkan tiga kelompok isolat. Kelompok I terdiri atas isolat Pleurotus sp.17, Pleurotus sp.16, Pleurotus sp.21, Pleurotus sp.27, dan Pleurotus sp.9, kelompok II terdiri atas isolat Pleurotus sp.4 dan Pleurotus sp.5, serta kelompok III yang hanya berisi satu isolat yaitu Pleurotus sp.24. Pengelompokan berdasarkan marka RAPD tersebut sejalan dengan karakteristik morfologinya. Informasi mengenai karakter morfologis dan genetik jamur tiram diharapkan akan bermanfaat untuk pengembangannya sebagai komoditas jamur komersial.Indonesian rainforest is one of the world's centers of biodiversity, which one of them is the oyster mushroom (Pleurotus spp.). This research was aimed to determine the morphological and genetic characteristics of eight isolates of Pleurotus spp.. The research was conducted from September 2005 to April 2006 at Forestry Pathology Laboratirum and Silvyculture Lab., and Forestry Biotechnology Lab. and Molecular Microbiology, Center of Biology Study, Agricultural Bogor Institute. The mushroom fruit body of the eight isolates was used as the material for observation of morphological and genetic characters. The results showed that the fruit body of eight isolates exhibited white, brown, or pink in color, with or without stalk, and full or half circle of cap shape. Amplification with RAPD primers OPO11 produced 12 bands, which one band was monomorphic while the others were polymorphic that showed the variability of the eight oyster mushroom isolates. Clustering based on banding patterns of amplification primers resulted in three groups. Group I consisted of Pleurotus sp.17, Pleurotus sp.16, Pleurotus sp.21, Pleurotus sp.27, and Pleurotus sp.9 isolates. Group II included Pleurotus sp.4 and Pleurotus sp.5 isolates, while the third group contained only one isolate i.e. Pleurotus sp.24. The molecular grouping was in line with the morphological characters. Information of morphological and genetic characteristics will hopefully give benefit for the development of the oyster mushroom as one of the commercial commodities.

Pengaruh Kombinasi Dosis Pupuk N, P, Dan K Terhadap Pertumbuhan Dan Hasil Tanaman Terung (Solanum Melongena L.) [the Influence of Dose Combination Fertilizer N, P, and K on Growth and Yield of Eggplant Crops (Solanum Melongena L.)]

Balanced fertilization is a basic requirement of success in improving the productivity of eggplant. One effort to looking proper dose. The purpose of this study was to determine the effect of the combination of fertilizer N, P, and K on the growth and yield of eggplant, and to get a dose of NPK fertilizers are best in improving productivity. This study was conducted in February – July 2013 in the experimental orchard of Indonesian Tropical Fruit Research Institute, Wera, Subang. The experimental design used was a randomized block design with 11 treatments and three replications consisting of (a) (control) 0 kg/ha, (b) 0 kg N/ha + 100 kg P2O5/ha + 75 kg K2O/ha, (c) 100 kg N/ha + 100 kg P2O5/ha + 75 kg K2O/ha, (d) 200 kg N/ha + 100 kg P2O5/ha + 75 kg K2O/ha, (e) 300 kg N/ha + 100 kg P2O5/ha + 75kg K2O/ha, (f) 100 kg N/ha + 0 kg P2O5/ha + 75 kg K2O/ha, (g) 100 kg N/ha + 200 kg P2O5/ha + 75 kg K2O/ha, (h) 100 kg N/ha + 300 kg P2O5/ha + 75 kg K2O/ha, (i) 100 kg N/ha + 100 kg P2O5/ha + 0 kg K2O/ha, (j) 100 kg N/ha + 100 kg P2O5/ha + 150 kg K2O/ha, and (k) 100 kg N/ha + 100 kg P2O5/ha + 225 kg K2O/ha. The results showed that NPK fertilization effect both on the growth and yield of eggplant, treatment NPK, fertilizer dose of 200 kg N/ha, 100 kg P2O5/ha, 75 kg K2O/ha give best effect on all parameters of observation. Implications of research singer has found the optimal dose that can be become the benchmark of fertilizer needs on eggplant.

Keragaman Jeruk Gunung Omeh (Citrus Nobilis Lour.) di Sumatera Barat Berdasarkan Marka RAPD [The Diversity Of Gunung Omeh Citrus (Citrus Nobilis Lour.) In West Sumatera Based On RAPD Marker]

The Gunung Omeh citrus (Citrus nobilis Lour.) is a local citrus growing throughout the citrus center of West Sumatera. However, the fruits available in the market are very diverse phenotypic. The purpose of this study is to classify the spread of citrus cv. Gn. Omeh derived from four districts based on genetic and morphological characters. The study was conducted from January to June 2016, with 21 of leaf and fruit samples collected from four Citrus District Regional Development farmers in the West Sumatera (Limapuluh Kota, Agam, Solok Selatan, and Tanah Datar). Besides that, control leaf samples derived from Budwood Multiplication Block/BPMT and Single Mother Tree/PIT were used. The morphological diversity both leaves and fruits were analyzed by Principal Component Analysis (PCA). The result showed that the level of genetic similarity in 23 samples of orange Gn. Omeh analyzed using two markers RAPD namely OPA 04 and OPA 018 which generate 24 bands, where 83.3% were polymorphic The resulting data were further analyzed by cluster analysis to observe groupings. Based on the dendrogram calculated according to UPGMA, 23 samples genetically are divided into two major groups. In the first group, there are two samples from Limapuluh Kota, while the remaining 21 samples are in second group. On the degree of similarity between 86.5–96%, PIT similar to the A1 plant and it belong to the same subgroup with S2, A5, T3, T5, S1. While BPMT similar to T4 plant, and it belong to the same subgroup with A4 and A3. Based on morphological characters at a 75% degree of similarity, the Gn. Omeh citrus in West Sumatra is divided into five groups, where the L1, T5, S5, and S2 plant sample belongs to the groups 1, 3, 4, and 5, respectively, while 18 others in Group 2. From the analysis of both genetic and morphological characters, it generates fairly high degree of variation among 23 samples. This shows that the diversity of the marketed is thought to be caused by the use of seed plants and crop cultivated management are diverse.
