Variasi Kandungan Kimia Minyak Cendana (Santalum Album Linn) dari Berbagai Provenans di Indonesia

Kualitas minyak cendana (Santalum album Linn) ditentukan oleh adanya 2 kandungan komponen Sesquiterpene, yakni α-santalol dan β-santalol. Tujuan penelitian ini untuk mengetahui variasi kandungan kimia cendana dari berbagai provenans. Sebanyak 54 sampel dari 8 provenans dan satu ras lahan diambil dari tanaman cendana umur 13 tahun di Hutan Penelitian Watusipat, Gunungkidul, Yogyakarta. Serbuk kayu cendana didapatkan dengan cara mengebor batang cendana setinggi 10 cm di atas permukaan tanah. Serbuk diekstrasi dengan larutan n-hexane untuk mendapatkan larutan minyak. Kandungan kimia larutan cendana dianalisis menggunakan GC-MS (gas chromatography-mass spectrometry). Hasil analisis menunjukkan adanya variasi kandungan kimia baik antar provenans maupun individu dalam provenans yang ditemukan pada 11 sampel dari 6 provenans. Senyawa golongan Sesquiterpene, Monoterpene dan komponen kimia lainnya ditemukan dalam jumlah yang bervariasi. Komponen utama dari hasil pengujian adalah α-santalol (32,38%-78,29%) dan β-santalol (5,45%-37,83%) diikuti oleh epi-β-santalol (5,97%-17,16%), P-Menth-2-En-9-OL-Trans (2,87%-8,53%) dan α-sinensal (3,83%-9,39%).

Effect of Media and Growth Regulators on the Propagation of Nyawai (Ficus Variegata Blume) Shoot Cutting)

Forest timber plantation of nyawai (Ficus variegate Blume) established by using vegetative propagated seedling from superior individu is very potential to develop for fulfilling the raw material demand of wood processing industries. The purpose of research was to get the optimum rooting medium and growth regulator concentration for rooting shoot cuttings of nyawai. Materials cuttings were provided from seedlings at 12 months age. The experiment was performed as factorial in a completely randomized design with two factors, i.e. rooting media factors (sand, mixed coconut coir dust + rice husk (2: 1, v/v), and mixed coconut coir dust+ husk rice + husk rice charcoal (6: 3: 1, v/v)) and IBA growth regulator concentrations (0, 50, 100,200,400, 600, and 800 mg/l), with three replications so that twenty experimented unit were used at each replication. Rooting percentage, root number, root length, shoot length, root biomass, shoot biomass, and leaf number were evaluated. Based on the result, sand media produced the highest rooted cuttings (88.10%), while the use of IBA showed the trend to increase the number of root with increasing the concentrations, but it influenced to reduce the shoot growth. Combination treatment of sand media and IBA 800mg/l provided the highest rooted cuttings (93.33%) with the number of root 21. Therefore, for the successful propagation of nyawai, cuttings are planted in sand media with IBA 800 mg/l.

Struktur Genetik Calliandra Calothyrsus Di Indonesia Menggunakan Penanda Random Amplified Polymorphism Dna (Rapd)

Calliandra calothyrsus was introduced to Indonesia from Guatemala in 1930s and had been planted widely throughout Indonesia for fuel wood. Genetic diversity within and among population is an important factor for initiating breeding program. Aim in this study was to evaluate genetic structure of C. calothyrsus in Indonesia, to obtain a better understanding of the history of the distribution and efficiency of breeding strategy of this species. Leaf samples were collected in a seedling seed orchard plot from 10 populations. Six RAPD markers consisted of 34 loci were used to analyze genetic diversity and genetic structure. The results showed that genetic diversity was in low to moderate level (mean HE= 0.321). The AMOVA analysis showed that genetic differentiation among geographical sources was significant, also among populations within geographical sources and among individual within populations as well. It was revealed that the 10 populations originated from only two ancestors. The limited number of ancestors resulted low to moderate genetic distances among populations (mean Da=0.070). In conclusion, C. calothyrsus has moderate genetic diversity within population and weak genetic structure among populations. Genetic diversity in population/individual level should be considered as a selection unit in the breeding strategies.
